Abstract

Catechins are flavonoid compounds that are the main components in gambier plants, where the largest gambier production in Indonesia comes from West Sumatra. Research on the depigmenting agent effect of gambir catechins has not been done much. The melanogenesis inhibitory effect of a depigmenting agent can be done by observing the decrease in the activity of the tyrosinase enzyme and the melanin content. This study aims to examine the activity of gambir catechins as depigmenting agents by testing the toxic effects of gambir catechins using the MTT assay, tyrosinase activity and melanin content as well as the expression of proteins involved in the mechanism of melanin formation in vitro using B16F0 cells. From the research conducted, it was found that gambier catechins affect the viability of B16F0 cells without causing toxicity to cells. Increasing the concentration and duration of the test significantly decreased cell viability (p<0.05). Gambier catechins have IC50 tyrosinase inhibition enzyme values which are almost the same as kojic acid, increasing the concentration and duration of the test significantly decreased tyrosinase activity and melanin content (p<0.05). Gambir catechins decreased the protein expression of CREB, β-catenin, ERK and MITF with a significant increase in concentration (p<0.05). Gambir catechins can inhibit melanogenesis in the cAMP signaling pathway. Based on the results of the study, it can be concluded that gambier catechins have an effect as a depigmenting agent.
