Abstract

Hot springs are natural habitats for thermophilic bacteria capable of producing thermostable protease enzymes. These enzymes have high economic value due to their wide range of applications in various industries, particularly those requiring high-temperature processes. Solok Regency has several hot springs, such as Garara, Padang Damar, and Batu Bajanjang, which have the potential to be habitats for these bacteria. This study aims to isolate, screen, and characterize some of the properties of thermophilic bacteria producing protease enzymes from these three locations. Isolation was performed at 50°C and yielded several isolates showing proteolytic activity. Some isolates with the highest proteolytic indices were selected for further characterization through macroscopic, microscopic, and biochemical analyses. Observations revealed that the isolates exhibited varying morphological and biochemical characteristics. One isolate demonstrated the highest specific activity at a specific harvest time, indicating its potential for industrial application. The results of this study reinforce that the hot springs of Solok Regency are a potential source for obtaining thermophilic bacterial isolates producing protease enzymes that are stable at high temperatures and have prospects for further development in the field of biotechnology.
