Abstract

The mesenchymal stem cell (MSC) secretome has potential as an active sunscreen agent by protecting skin fibroblasts from photoaging induced by UVB radiation. The MSC secretome is a secretion product from MSC cultures containing bioactive compounds such as cytokines, proteins, and growth factors with regenerative and antioxidant potential. This study aimed to evaluate the antioxidant and sunscreen activity of formulations containing MSC secretome. The secretome was obtained from the culture medium of adipose-derived MSCs using DMEM at passage 5. The sunscreen was formulated in three concentrations of secretome: 8% (F1), 10% (F2), 12% (F3), and a base formula. The evaluated sunscreen formulations resulted in homogeneous emulsions, white in color, with a characteristic aroma, pH ranging from 6.24 to 6.74, of M/O type, with thixotropic flow properties, and stable under freeze–thaw testing. Antioxidant activity was determined using the FRAP method with ascorbic acid as the positive control, and SPF values were measured in vitro using the Mansur method. FRAP results for the base, F1, F2, F3, pure secretome, and ascorbic acid were 0.895; 0.838; 0.854; 0.854; 0.840; and 7.635 µmol Fe²⁺/mg, respectively. Statistical analysis showed no significant difference in FRAP values between the secretome-containing formulas and the base (p > 0.05). SPF values for F1, F2, F3, and the base were 12.410; 15.748; 2.406; and 1.280, respectively (p < 0.05). The results indicate that the sunscreen formulation containing MSC secretome exhibits sunscreen activity, with F2 achieving the highest SPF value categorized as ultra protection.
