Abstract

Introduction: Muhammad Iqbal, M. Iqbal Rivai, Rini Suswita Abstract Environmental and genetic factors, particularly the gut microbiota, contribute to colorectal cancer (CRC) development. This study explores the relationship between gut microbiota changes and CRC occurrence, development, prognosis, and early detection. Fecal samples are commonly used for non-invasive analysis, allowing effective screening for CRC risk. Methods: A laboratory experimental research design utilizing Sprague Dawley rats induced with azoxymethane (AOM) and Dextran Sodium Sulfate (DSS) was conducted. Thirty rats were categorized into normal, cancer-induced, and treatment groups. Fecal microbiota profiles were analyzed using Next Generation Sequencing (NGS). Data were processed for alpha and beta diversity, revealing microbial community dynamics. Results: Sequencing results confirmed validity, exhibiting varying sequence reads in normal, cancer-induced, and treated rats. Firmicutes dominated at the phylum level, with Bacilli and Clostridia prevalent at the class level. Order-level analysis demonstrated uneven bacterial abundances among samples. Alpha diversity indicated lower diversity in treated rats, while beta diversity revealed compositional similarities between treated and normal rats, distinguishing them from cancer- induced rats. Conclusion: The study provides valuable insights into gut microbiota dynamics in CRC models. Firmicutes dominance, specifically Bacilli and Clostridia classes, underscores microbial composition. Uneven bacterial abundances at different taxonomic levels and reduced alpha diversity in treated rats suggest a potential probiotic effect of Lactococcus lactis D4 in preventing dysbiosis. Beta diversity highlights compositional similarities between treated and normal rats, emphasizing the potential of Lactococcus lactis D4 in modulating gut microbiota composition and mitigating CRC risk.
