Abstract

Piperin merupakan salah satu senyawa utama dari tanaman lada hitam (Piper ningrum). Zat ini memiliki banyak efek farmakologis, yaitu sebagai antiinflamasi, analgesik, antidepresan, antileukemia, antioksidan dan sitoprotektif. Piperin memiliki kelarutan yang rendah didalam air, hal ini menyebabkan bioavaibilitasnya juga rendah sehingga diperlukan strategi untuk dapat meningkatkan kelarutan dan bioavaibilitasnya. Tujuan dari penelitian ini adalah untuk membentuk sistem biner piperin dengan asam malonat dan trometamin sebagai koformer untuk meningkatkan kelarutan dari piperin. Metode yang digunakan didalam penelitian ini adalah solvent evaporation yang bertujuan membentuk sistem biner antara piperin-asam malonat dan piperin-trometamin. Dalam penelitian ini dibuat sistem biner piperin-asam malonat dengan perbandingan equimolar 1:1 dan 2:1, sistem biner piperin-trometamin dengan perbandingan equimolar 1:1, dan 2:1. Sampel ini dikarakterisasi dengan analisa difraksi sinar X (XRD), differential scanning calorimetry (DSC), dan fourier transform infrared spectroscopy (FTIR). Dari hasil difraksi XRD menunjukan berkurangnya intensitas puncak interferensi yang dihasilkan. Dari hasil analisa DSC didapatkan bahwa entalpi energi menurun. Pada Uji FTIR terdapat pergeseran spektrum yang mengindikasikan terbentuknya ikatan hidrogen. Peningkatan kelarutan tertinggi ada pada formula sistem biner piperin-trometamin 1:1 dengan peningkatan kelarutan 3,95 kali dari piperin murni. Peningkatan disolusi tertinggi ada pada formula sistem biner piperin-trometamin 1:1 dengan kadar terdisolusi 68,97%. Kata kunci : piperin, asam malonat, trometamin, sistem biner, solvent evaporation, kelarutan FORMATION THE BINARY SISTEMS OF PIPERINE-MALONIC ACID AND PIPERINE-TROMETHAMINE USING SOLVENT EVAPORATION METHOD ABSTRACT Piperine is one of the main compounds of the black pepper plant (Piper ningrum). This substance has many pharmacological effects, namely as an anti-inflammatory, analgesic, antidepressant, antileukemia, antioxidant and cytoprotective. Piperin has a low solubility in water, this also causes low bioavailability so a strategy is needed to increase solubility and bioavailability. The aim of this study was to prepare binary system of piperine using malonic acid and tromethamine as coformers in order to increase solubility of piperin by solvent evaporation technic. In this study the binary systems of piperine-malonic acid was prepared with a ratio of equimolar 1:1 and 2:1, and the binary sistems of piperine-trometamine was prepared with a ratio of equimolar 1:1, and 2:1. This sample was characterized by X-ray diffraction analysis (XRD), differential scanning calorimetry (DSC), and fourier transform infrared spectroscopy (FTIR). Based on the results of XRD diffraction showed the decrease intensity of the interference peak produced. The DSC analysis found that the energy enthalpy decreased. The FTIR result showed a shift in the spectrum which indicated the formation of hydrogen bonds. The highest increase in solubility was found in the 1:1 piperine-trometamine binary sistem formula with an increase in solubility of 3.95 higher than intact piperine. The highest increase in dissolution was also found in the 1:1 piperine-trometamine binary system formula with an increase in dissolution profile by 68.97%.
