Abstract
Solid dispersion is used to increase the solubility of atorvastatin calcium which has low solubility in water. The solid dispersion methods used in this study were solvent evaporation method and spray drying. Atorvastatin calcium-polyvinylpyrrolidone K-30 (PVP K-30) solid dispersion as a polymer was prepared in the ratio of 1:9, 3:7, 5:5, 7:3, and 9:1. The best solid dispersion solubility and dissolution test results were characterised by scanning electron microscope (SEM), powder x-ray diffraction (PXRD), differential scanning calorimetry (DSC) and fourier transform infra red (FTIR) analysis. Solvent evaporation method and spray drying have the best solubility at a ratio of 1:9 with an increase in solubility of 9.98 times and 11.12 times, respectively. The results of the best percent dissolved solid dispersion of solvent evaporation method and spray drying at a ratio of 1:9 with percent dissolved at the 30th minute in phosphate buffer Ph 6.8 were 86.136% and 92.870%, respectively. Based on the characterisation results, atorvastatin calcium and polyvinylpyrrolidone K-30 (PVP K-30) solid dispersion was formed, which is amorphous. In conclusion, the solid dispersion of atorvastatin calcium and polyvinylpyrrolidone K-30 (PVP K-30) using solvent evaporation method and spray drying can increase the solubility of atorvastatin calcium.
