Abstract
The research on the isolation and screening of thermophilic bacteria producing amylase enzymes from the Sipoholon-Tarutung geothermal region has been carried out at the Microbiology Research Laboratory, Department of Biology, Andalas University from December 2018 to March 2019. Bacteria are isolated from hot springs at Sipoholon-Tarutung, North Sumatra. This study aims to obtain amylolytic thermophilic bacteria, analyze amylolytics and analyze their characterization. This study used a purposive sampling method based on gradient temperatures in hot springs which ranged from 500C to 600C. The results were obtained by 5 amylolytic isolates from thermophilic bacteria Sipoholon-Tarutung hot springs with amylolytic indices ranging from 0.30 to 2.30, isolate BTSA-01 has the highest amyloytic index and amylase enzyme producing potential. Characterization of macroscopic, microscopic and biochemical tests was reported. These isolates were characterized as aerobic and Gram positive bacteria.
