Abstract

Piperin is one of the major compound of black pepper plants (Piper ningrum). This compound has many pharmacological effects, namely as an anti-inflammatory, analgesic, anti-depressant, anti-leukemia, anti-oxidant and citoprotective. Piperine has low solubility in water, this causes its bioavailability also low, so a strategy is needed to increase its solubility and bioavailability. The method used in this study is the melting of solid dispersions with polymer Gelucire 44/14. In this research piperine-Gelucire 44/14 solid dispersion with 3 ratio (1:1, 2:1; 1:2) and physical mixture (1:1). Solid dispersion and piperine-Gelucire 44/14 physical mixture were characterized by XRD diffraction analysis, differential scanning calorimetry (DSC), scanning electron microscopy (SEM) and fourier transform infrared spectroscopy (FTIR). From the results of X-ray diffraction shows the reduced intensity of the peak interference produced. From the results of DSC analysis it was found that the peak intensity was reduced. The SEM test showed that piperine crystals were dispersed in gelucire 44/14 so that the solid dispersion was in amorphous form. In the FTIR test the peak, Indicates the formation of hydrogen bonds. This bond prevents the recrystallization process from piperine. The highest solubility increase was in the 1:2 solid dispersion formula with an increase in solubility of 3.32 higher than piperine. The highest dissolution increase was in formula 1:2 with 4.03 dissolution efficiency increase from piperine.
