Abstract

Latar belakang : Streptococcus mutans merupakan flora normal yang terdapat di rongga mulut. Bakteri ini merupakan bakteri gram positif yang bersifat anaerob fakultatif dan dapat menghasilkan asam yang dapat mengakibatkan demineralisasi email gigi. Rimpang kunyit (Curcuma longa) adalah tanaman obat yang memiliki aktivitas antibakteri karena mengandung senyawa kurkumin. Kurkumin merupakan senyawa fenolik yang dapat menghambat pertumbuhan bakteri dengan cara mendenaturasi dan merusak membran sel yang dapat mengakibatkan kematian sel. Kurkumin bekerja dengan cara menginaktivasi enzim sehingga dapa menganggu kelangsungan pertumbuhan bakteri. Tujuan : Tujuan dari penelitian ini adalah untuk mengetahui daya hambat ekstrak rimpang kunyit berbagai konsentrasi terhadap bakteri Streptococcus mutans secara in-vitro. Metode : metode penelitian ini adalah eksperimental laboratorium dengan desain penelitiannya adalah post test only control group design. Sampel yang digunakan yaitu rimpang kunyit yang berasal dari ladang kunyit puncak payo, Tanah Garam Solok. Penelitian dilakukan di Laboratorium Biota Universitas Andalas dan Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas. Penelitian tahap awal dilakukan dengan membuat ekstrak rimpang kunyit konsentrasi 20%, 40%, 60%, 80%, dan 100%. Kemudian dilanjutkan dengan pengujian daya hambat ekstrak dengan metode difusi menggunakan cakram. Zona bening yang terbentuk pada MHA di cawan petri diukur dengan menggunakan jangka sorong digital. Analisis data yang digunakan adalah uji Kruskal Wallis dan dilanjutkan dengan uji Mann-Whithney. Hasil : hasil penelitian didapatkan bahwa ekstrak rimpang kunyit (Curcuma longa) konsentrasi 20% dan 40% tidak memiliki daya hambat namun konsentrasi 60%, 80%, dan 100% memiliki daya hambat Kesimpulan : Ekstrak rimpang kunyit (Curcuma longa) konsentrasi 20% dan 40% tidak memiliki daya hambat sedangkan ekstrak rimpang kunyit (Curcuma longa) konsentrasi 60%, 80% dan 100% memiliki daya hambat terhadap bakteri Streptococcus mutans dan ekstrak rimpang kunyit (Curcuma longa) berbagai konsentrasi belum cukup efektif dalam menghambat pertumbuhan Streptococcus mutans. Kata kunci : Curcuma longa, daya hambat, difusi, dan Streptococcus mutans Test Of Turmeric (Curcuma longa) Extract In Various Concentration On Inhibiting Streptococcus mutans ATCC 31987 In In-Vitro Grace H. Sihombing, Linosefa, Hidayati Background: Streptococus mutans was a normal flora in oral cavity, gram positive cocci, anaerobic facultative, and could produce acid that cause demineralization of tooth enamel. Turmeric was a medicinal plant that has antibacterial activity because it contained curcumin. Curcumin was a phenolic compound that can denature and destroy cell membranes. Curcumin worked by inactivating the enzyme so that it could interfere with the growth of bacteria. Purpose : The purpose of this study was to determine the inhibitory extracts of turmeric various concentrations of Streptococcus mutans bacteria in-vitro. Methods : the research method was experimental laboratories with posttest only control group design. Samples were used in this research are turmeric rhizome took from turmeric field, Tanah Garam Solok. This research was conducted at Andalas University Biota Laboratory and Microbiology Laboratory Faculty of Medicine Andalas University. Early stage was done by making extracts of turmeric with concentrations are 20%, 40%, 60%, 80%, and 100%. Then, the extracts was tested into the bacteria with the diffusion method used disk. The clear zone formed on the MHA in the petri dish were calculated using calipers. Data was analyzed by using Kruskal Wallis test and continued with Mann-Whithney test. Results: The result presented that 20% and 40% turmeric exctract didn’t have inhibition activity but 60%, 80%, and 100% turmeric exctract had inhibition activity. Conclusions: 20% and 40% turmeric extract didn’t have inhibition activity but 60%, 80%, and 100% turmeric exctract had inhibition activity and various concentration of turmeric extract have not been quite effective on inhibiting the growth of Streptococcus mutans Keyword(s) : Curcuma longa, diffusion, Streptococcus mutans, and inhibition activity.
