Abstract
Legundi plant (Vitex trifolia L.) is a medicinal plant that belongs to the Lamiaceae family which is classified as a bush plant. One part of the legundi plant that is commonly used as traditional medicine is their leaves. Leaves of the legundi plant are commonly used to reduce pain, dizziness, colds, reduce heat, relieve spasms, coughs, tonsillitis, tuberculosis and typhus.This study aims to determine the phytochemical profile, total phenolic content, antioxidant activity, and antibacterial properties of the metanol extract, hexane fraction, ethyl acetate fraction, and butanol fraction of legundi leaves. The results of the phytochemical profile test showed that legundi leaf extract and fractions had secondary metabolite compounds in the form of phenolics, flavonoids, tepernoids, steroids, and saponins. Determination of total phenolic content was carried out using the Folin-Ciocallteu method with the highest concentration in the ethyl acetate fraction (55.2 mg GAE/g dry fraction). The antioxidant activity test was carried out using the DPPH method, the results showed that the butanol fraction had the strongest activity against free radicals with an IC50 value (12.99 mg/L). The antibacterial test was carried out using the disc diffusion method which showed that the butanol fraction had the greatest ability to inhibit bacterial growth with an inhibition zone for Staphyloccocus aureus (11.26 mm) and Escherichia coli (7.753 mm).
