Abstract

Kombinasi prednison-NaCl pada tikus putih digunakan untuk menginduksi kondisi preeklampsi. Kombinasi ini digunakan selama trimester II-III kehamilan. Sebanyak 12 ekor tikus hamil digunakan (dibagi menjadi 4 kelompok). Kelompok 1 sebagai kontrol hamil, kelompok 2, 3 dan 4 diinduksikan prednison 2,5 mg/kg dan NaCl yang divariasikan 200, 400 dan 800 mg/kg. Campuran ini diberikan satu kali sehari selama trimester II-III kehamilan (hari 11 s/d 21). Tekanan darah sistol, diastol, tekanan arteri rata-rata dan denyut jantung diukur setiap dua hari sekali selama kehamilan dan setelah melahirkan menggunakan pengukuran secara tidak langsung (CODA®). Pengamatan kadar protein urin dilakukan pada hari ke-20 kehamilan. Setalah kehamilan dilakukan pengamatan terhadap jumlah anak dan berat anak lahir. Analisis data dilakukan dengan menggunakan ANOVA dua arah yang dilanjutkan dengan Duncan untuk semua parameter kecuali jumlah anak dan berat anak menggunakan ANOVA satu arah. Berdasarkan hasil analisis data dan pengamatan diketahui bahwa Tekanan darah sistol, diastol, tekanan arteri rata-rata dan denyut jantung kelompok dosis uji lebih tinggi (p<0.05) dibandingkan dengan kelompok kontrol. Jumlah anak dan berat anak lahir kelompok kontrol lebih tinggi (p<0.05) dibandingkan dengan kelompok uji. Kadar protein urin pada dosis uji lebih tinggi dibandingkan dengan kelompok kontrol. Hal ini menunjukkan model hewan preeklampsi dengan menggunakan induksi prednison-NaCl pada trimester II-III bisa digunakan pada tingkatan dosis prednison 2,5 mg/kg-NaCl 400 dan 800 mg/kg. ANIMAL MODEL PREECLAMPSIA USING PREDNISONE-NaCl INDUCTION IN 2ND -3 RD TRIMESTER ABSTRACT The combination of prednisone-NaCl to induce preeclampsia conditions on pregnant rat have been conducted. This combination was given once a day during 2nd - 3 rd trimesters of the pregnancy (days 11 to 21). Animal consist of 12 pregnant rat were used (divided into 4 group). Group 1 as control, group 2,3 and 4 treated with 2,5 mg/kg prednison and 200, 400 and 800 mg/kg NaCl. Systolic blood pressure, diastole, mean arterial pressure and heart rate were measured every two days and after delivery using non-invasive instruments (CODA®). Number of offspring, their body weight and mother urine protein were also determined. Data was analyzed by two-way ANOVA followed by Duncan Multiple Range Test for all parameters except the number of offspring and their body weight using one-way ANOVA. Significance was taken at (p<0.05). Based on the results of data analysis and observation it is known that systolic blood pressure, diastole, mean arterial pressure and heart rate of prednison-NaCl treated rat were higher (p <0.05) compared to the control group. The number of offspring and their bodyweight of prednison-NaCl treated rat were lower (p <0.05) compared to control group. Urine protein levels at treated rat were higher than in the control group. Theese indicated that preeclampsia can be induced by combination of prednisone-NaCl during 2nd -3 rd trimesters of the pregnancy using prednisone-NaCl induction at 2.5 mg/kg prednisone and 400-800 mg/kg. .
