Abstract
Alzheimer's disease is a chronic progressive neurodegenerative disorder that most people in the world suffer from. In several neurodegenerative diseases such as Alzheimer's, stem cell therapy is a promising treatment opportunity. This research aims to study the effect of administering mesenchymal stem cells derived from adipose, bone marrow and Wharton Jelly on the expression of the Nestin, SOX2 genes and the appearance of amyloid plaques in Alzheimer's rats. This research is an experimental research with a Posttest-Only Control Group Design. The sample consisted of 10 AD rats as the control group and 15 AD rats treated with mesenchymal stem cells derived from adipose, bone marrow and Wharton Jelly as the treatment group. AD induction was carried out by oral ingestion at a dose of 300 mg/kg BW of rats for 5 days in 1 ml of distilled water/100 kg BW of rats. Mesenchymal stem cells were administered via intra-peritoneal injection at a dose of 1x106 in 300 ul of complete medium. Then the rats brain tissue was harvested one month after stem cells injection and prepared for examination of Nestin, SOX2 gene expression and histopathological examination. The statistical test used was the ANOVA test for the expression of the Nestin, SOX2 genes and the appearance of amyloid plaques. Nestin gene expression was higher in the group treated with AD-MSCs compared to the positive control group. SOX2 gene expression was higher in the group treated with BM-MSCs compared to the positive control group. The appearance of amyloid plaques in the rats brain cortex tissue was lowest in the group treated with WJ-MSCs compared to the positive control group. The appearance of amyloid plaques in the hippocampus tissue of the rat brain was lowest in the group treated with AD-MSCs compared to the positive control group. This study concluded that there was an increase in Nestin and SOX2 gene expression after administering mesenchymal stem cells derived from adipose, bone marrow, Wharton Jelly to Alzheimer's rats. There was a decrease in the appearance of amyloid plaques in the cortex and hippocampus after administration of mesenchymal stem cells derived from Wharton Jelly, adipose, and bone marrow in Alzheimer's rats.
