Abstract

Indarung (Trema orientalis (L.) Blume) is a pioneer plant species that has the ability to reclamate the mine wastelands. Although Indarung is a good candidate for reforestation, low percentage and nonsynchronous germination prevents this species from being efficiently used for large scale reforestation programs. Tissue culture technique is one of solution for propagating of Indarung using seed as source of explants. Furthermore, by using cytokinins, it will be supported the effectiveness of seed germination. The aims of the research were to evaluate several cytokinins and determine the best cytokinins for initiate of Indarung seed germination on WPM medium. The research used a Completely Randomized Design (CRD) with four treatments and six replications. The treatments were (A) WPM without cytokinins as control, (B) BAP 0.7 mg.L-1, (C) KIN 4 mg.L-1, and (D) TDZ 0.1 mg.L-1. The germination were observed for 60 days. The results showed that 0.7 mg.L-1 BAP was the best cytokinin for seed germination of Indarung. Meanwhile, 4 mg.L-1 KIN showed the best result on the first day of root appeared, length of roots, height of shoots, and morphology of shoot. Plantlet of Indarung on WPM media supplemented with 4 mg.L-1 KIN have a long branched roots and greeny leaves with dark green pointed tip.
