Abstract
ABSTRACT THE EFFECT OF Lactobacillus crispatus BACTERIOCIN TEST ON INHIBITING GROWTH OF Staphylococcus aureus By Mella Warizka Lactobacillus crispatus is a lactic acid bacteria producing as an antibacterial agent. This reserch aims to find the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) of bacteriocin filtrate from L. crispatus on inhibiting Staphylococcus aureus growth in vitro. This research was conducted at the Microbiology Laboratory of the Andalas University Faculty of Medicine in May 2019-January 2020. The population of this study was S. aureus and the study sample was S. aureus which had been isolated in the Microbiology Laboratory of the Faculty of Medicine, Andalas University. This research was done using dilution technique with completely randomized design. The bacteriocin filtrates of L. crispatus were divided into several concentration; 100%, 80%, 60%, 40%, 20%, and 0% (control). MIC and MBC were determined by counting the number of growing bacterial colonies in each concentration. The data was statistically analyzed by Kruskal Wallis test and Mann Whitney test. The study showed that filtrate of L. crispatus had an inhibitory effect on the growth of S. aureus. The MIC and MBC from bacteriocin of L. crispatus cannot be identified. This research concludes that bacteriocin filtrate of L. crispatus has bacteriostatic effect on S. aureus without no bactericidal activity. The antibacterial ability of L. crispatus against the growth of S. aureus depends on the concentration of bacteriocin filtrate. Keywords: Antibacterial, bacteriocin, Lactobacillus crispatus, Staphylococcus aureus ABSTRAK UJI DAYA HAMBAT BAKTERIOSIN Lactobacillus crispatus TERHADAP PERTUMBUHAN Staphylococcus aureus Oleh Mella Warizka Lactobacillus crispatus adalah salah satu jenis bakteri asam laktat yang dapat menghasilkan bakteriosin sebagai suatu senyawa antibakteri. Penelitian ini bertujuan untuk mengetahui Kadar Hambat Minimal (KHM) dan Kadar Bunuh Minimal (KBM) dari filtrat bakteriosin L. crispatus dalam menghambat pertumbuhan Staphylococcus aureus secara in vitro. Penelitian ini dilakukan di Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas pada bulan Mei 2019-Januari 2020. Populasi penelitian ini adalah S. aureus dan sampel penelitian adalah S. aureus yang telah diisolasi di Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas. Penelitian daya hambat filtrat bakteriosin L. crispatus terhadap S. aureus dilakukan dengan teknik dilusi dan desain penelitian Rancangan Acak Lengkap. Filtrat bakteriosin dibagi dalam berbagai konsentrasi yaitu 100%, 80%, 60%, 40%, 20%, dan 0% (kontrol). Nilai KHM dan KBM ditentukan dengan menghitung jumlah koloni bakteri yang tumbuh dari masing-masing konsentrasi. Hasil penelitian dianalisis secara statistik dengan uji Kruskal Wallis dan Mann Whitney. Hasil penelitian menunjukkan filtrat L. crispatus dapat menghambat pertumbuhan S. aureus. Nilai KHM dan KBM filtrat bakteriosin L. crispatus tidak dapat diidentifikasi. Dari penelitian diperoleh hasil bahwa filtrat bakteriosin L. crispatus memiliki efek bakteriostatik terhadap pertumbuhan S. aureus tetapi tidak memiliki efek bakterisidal. Kemampuan daya hambat bakeriosin L. crispatus terhadap pertumbuhan S. aureus dipengaruhi oleh konsentrasi filtrat bakteriosinnya.
