Abstract

The fig plant (Ficus carica Linn) is widely used as traditional medicine by Middle Eastern people such as digestive medicine, itching, and inflammation. This study aims to determine the secondary metabolite compounds, cytotoxic activity and antibacterial activity of methanol extract and each leaf fraction. Extraction is carried out directly using methanol solvent which then the result of thick extract will be fractionated using hexane, ethyl acetate, butanol, and water. Fractionation is carried out in sequence according to the level of polarity, starting from non-polar to polar solvents. Cytotoxic activity was carried out by the Brine Shrimp Lethality Test (BSLT) method. Antibacterial activity is carried out by the disc diffusion method. The results showed that the methanol extract contained secondary metabolites of steroid alkaloids and triterpenoids, hexane fractions contained steroids and triterpenoids, ethyl acetate fractions contained steroids and triterpenoids, ethyl fractions contained steroids and triterpenoids and the remaining fraction (water) only contained saponins. Cytotoxic activity of methanol extract and each active fraction as a toxic compound with hexane fraction had the lowest LC50 value of 162,53,95 mg/L. The antibacterial activity test of methanol extract and each fraction showed the moderate activity of the bacteria against Staphylococcus aureus and Escherichia coli.
