Abstract
Many degenerative diseases are caused by free radical reactions. One of chemical compounds as the source of free radical is carbon tetrachloride (CCl4) which can affect body organs. But the major effect is in the liver as the metabolic process occurs in endoplasmic reticulum by cytochrome P450. The metabolized CCl4 can cause hepatocyt which degenerates hydropic and lipid and cause necrosis. The bad impact of CCl4 can be overcome by antioxidant. Roselle is a rich antioxidant plant, which also assumes has characteristic of hepatoprotector. It will protect hepatocytes from free radicals. This research is aimed to find out the effect of rosella extract treatment to cytochrome P450 CYP24A1 and induced CCl4 degenerated wistar rat liver cells. This research was an experimental study using the post test only controlled grou design. The samples were 24 wistar rats age 2 – 3 months old and weigh 200 – 250 g. They were randomly divided into 4 groups which are negative control group, positive control group (induced by CCl4), treatment I group ( induced by CCl4 and was given 250 mg/kg BW roselle extract) and treatment II group (induced by CCl4 and was given 500 mg/kg BW roselle extract). CCl4 was given orally in single dosage 1 mg/kg/BW 24 hours. Roselle extract was given after CCl4 for 6 days. In the 8th day, those rats were terminated for their blood and liver tissues. The blood was centrifuged to separate serum from other blood components. The serum was used to investigate cytochrome P450 CYP24A1 activities by using Elisa Method. Liver tissues were processed and made into slides and colored .Those slides were read under microscope. Data were analyzed by ANOVA test with 95% degree of trust. The result of the research has shown that cytochrome P450 CYP24A1 activities in negative control group 6.61±5.47 nmol/ml , and in positive control group increase 4.36 times, that is 28.83±8.42 nmol/ml and in treatment group 5.49±4.87 nmol/ml and treatment II group 1.50±0.84 nmol/ml. Statistically, it was found out that the differences average cytochrome P450 CYP24A1 activities among groups (p.The hydropic degeneration average of negative control group was 0.00±0.00, positive control group was 35.83±2.75 and treatment I group was 13.00±2.50, treatment II group was 2.44±1.07. Statistically, it was found out the difference of the average of hydropic degeneration among groups (p. The lipid degeneration average of negative control group was 0.17±2.85 and positive control group was 11.77±4.06, treatment I group was 7.78±2.85 and treatment II group was 4.28±1.00.Statistically, it was found out the difference of lipid degeneration among groups (p.
