Abstract
Daemonorops draco is a dioecious rattan species that produces highly valuable resin. Sex identification at the seedling stage is challenging due to the absence of secondary sexual characteristics, and flowering occurs only after 5–7 years. This study was conducted from September 2024 to July 2025 at the Genetic and Biomolecular Laboratory, Andalas University, using samples collected from Sako Besar Village and Bukit Duabelas National Park, Jambi. The aim was to identify ISSR primers capable of producing sex-specific DNA bands in Daemonorops draco, a dioecious species lacking early visible sexual characteristics. DNA was isolated from 15 male and 15 female individuals, assessed via NanoDrop, and amplified using 18 ISSR primers. All primers amplified DNA successfully, but only RT 28 produced an approximately 190 bp band exclusively in male individuals. The results indicate that RT 28 is a promising male-specific molecular marker for early sex identification, supporting conservation and cultivation programs, and is recommended for further development into a SCAR marker.
