Abstract

Daemonorops draco (Willd.) Blume, or dragon's blood rattan, is a dioecious plant that produces resin of high economic value. The cultivation and conservation of this species face challenges due to the difficulty in distinguishing between male and female individuals before they reach reproductive age, whereas only female individuals produce resin-yielding fruits. Molecular approaches such as Random Amplified Polymorphic DNA (RAPD) serve as an initial step for the identification of the sex of dioecious plants. This study aims to select RAPD primers capable of producing sex-specific bands in D. draco. The samples consisted of 15 male and 15 female individuals whose DNA was isolated using the modified CTAB protocol, and then amplified with 28 RAPD primers selected based on their success in sex identification in previous research. The results showed that 26 out of 28 primers successfully amplified D. draco DNA, producing bands with varying numbers (5–41 bands). Four primers produced sex-specific bands, namely OPA-02, OPC-19, OPD-10, and OPA-14. Two of these four primers, OPA-14 (455 bp) and OPC-19 (310 bp), showed male-specific bands that consistently appeared in all male individuals. Thus, these primers are strong candidates for development into SCAR (Sequence Characterized Amplified Region) sex markers because they are specific, stable, and reproducible.
