Abstract

This research was conducted to 1) identify the presence of endophytic fungi from the bark of Taxus sumatrana originating from Mount Singgalang, 2) describe the characteristics and growth potential of endophytic fungal isolates in vitro, 3) analyze the ability of the identified endophytic fungal isolates to produce Taxol compounds, and 4) molecularly identify potential Taxol-producing endophytic fungal species. The samples used in this study were bark tissues of Taxus sumatrana collected from Mount Singgalang at an altitude of 1559 meters above sea level. Based on morphotype identification results, Taxus sumatrana from Mount Singgalang was found to be associated with two species of endophytic fungi. The endophytic fungi isolated from the bark tissues of Taxus sumatrana, identified molecularly as TX01, showed 99.81% homology with Neopestalotiopsis sp., while TX02 showed 99.81% homology with Pestalotiopsis microspora. In vitro testing results indicated that the isolates grew well on PDA medium, with Neopestalotiopsis sp. optimal on sucrose as a carbon source and Pestalotiopsis microspora optimal on glucose in fermentation medium. Fermentation conducted for 21 days and analysis using HPLC confirmed that isolate TX01 produced Taxol compounds at a concentration of 2499 µg/Liter, while isolate TX02 produced Taxol compounds at a concentration of 365.14 µg/Liter.
