Abstract

The high demand for skins and body parts of Aonyx cinereus has led to frequent illegal wildlife trade make this species listed in Appendix I and IUCN Redlist Data as vulnerable species. Identification of morphological differences in illegal otter trade is difficult to obtain and may affect conservation policy for this species. Therefore, an accurate level identification method such as DNA barcoding is necessary for species-level identification. The objective of this study is to determine DNA barcode of Aonyx cinereus at Kayu Tanam in West Sumatra using Cytochrome Oxidase Sub unit I (COI) gene. The study was conducted from June to September 2023 using two fecal sample from Kayu Tanam, Padang Pariaman Regency. Based on COI gene fragment length along 590 bp nucleotide base, revealing that two samples of Aonyx cinereus Kayu Tanam has two specific base at 281 (T bases) and 581 (A bases) as marker identity of Aonyx cinereus Kayu Tanam. Based on phylogenetic analysis, we found that Aonyx cinereus kayu tanam and Sarawak closely related with genetic distance 0 – 0.7% and also has specific base at 32 (A bases) as marker identity Aonyx cinereus Sundaland.
