Abstract
Breast cancer is a disease characterized by the formation of a lump in the breast by a group of abnormal cells that continue to multiply. Current breast cancer treatment is still not optimal because it can also damage normal cells. One of the targets of anticancer therapy is to induce apoptosis. Metabolite compounds in sungkai leaf extract have anticancer potential. The purpose of this study was to analyze the effect of sungkai leaf extract on breast cancer cell viability and apoptosis and evaluate BCL2 gene expression. MTT assay was used to determine the IC50 value with control and treatment groups (48 and 72 hours) given ethanol extract of sungkai leaves at various concentrations. Molecular apoptosis assay using qPCR to see the expression of BCL2 gene. DAPI staining was used to observe the morphology of apoptotic cells through fluorescent microscopy. The results of cell morphology seen with an inverted microscope with IC50 decreased cell density and changes in cell shape. The results of the MTT test at 48 hours incubation obtained an IC50 value of 8.128 μg/ml, while at 72 hours incubation obtained an IC50 value of 0.339 μg/ml, indicating a very active cytotoxic effect. There was a decrease in BCL2 gene expression, at 48 hours showing statistical significance p = 0.0005 (p <0.05), while at 72 hours it was not significant (p>0.05). In the results of fluorescent microscopy, it can be seen that cells have undergone apoptosis, there is blue luminescence and fragmentation of cell DNA. Based on the results obtained, it can be concluded that sungkai leaf extract has anticancer potential by reducing cell viability, reducing BCL2 gene expression and visible cell morphology that experiences apoptsois in breast cancer cells.
