Abstract

Dendrobium mussauense Ormerod has beautiful flowers with a faint yellow color and has a high economic value. D. mussauense facin a high risk of becoming extinct in the wild. Somatic embryogenesis (SE) is a rapid vegetative propagation technique for the conservation of D. mussauense. The research aimed to evaluate the effects of types and concentrations of plant growth regulator (PGRs) combinations, namely 2,4-D (2,4-dichlorophenoxyacetic acid) with BAP (6-benzylaminopurine) and 2,4-D with TDZ (thidiazuron) on SE induction. Nodal stem segments of D. mussauense were cultured on a half-strength MS medium supplemented with 2,4-D alone, 2,4-D + BAP and 2,4-D + TDZ with different concentrations. The research used a Completely Randomized Design (CRD) with seven treatments and four replications. Results showed that the addition of 2,4-D alone or combinations of 2,4-D with BAP or TDZ provided significant differences in the emergence time of SE, but there were no significant differences in the percentage of SE and number of SE in D. mussauense. The combination of 2,4-D and TDZ to the medium resulted in the shortest time for SE emergence than 2,4-D alone. The most effective treatment, 2,4-D 1 mg/L+ 0.25 mg/L of TDZ was the shortest time for SE emergence.
