Abstract
Breast cancer and cervical cancer persists as a critical health concern. The multifaceted side effects associated with cancer therapies underscore the urgent need for alternative modalities with enhanced tolerability necessitating the exploration of alternative therapeutic with reduced side effects compared to conventional treatment. Apoptosis, a programed cell death, emerges as one of effective target for cancer therapy. This research delves into the potential of Java cardamom seed, a natural product rich in active anticancer constituents, as a therapy for breast and cervical cancer. The primary objective is to ascertain the impact Java cardamom seed extract on the cancer cell viability and elucidate underlying mechanism of apoptosis. Java cardamom seeds were subjected to a maceration employing 96% ethanol solvent. Identification of secondary metabolites was accomplished through phytochemical test. Subsequently, T47D cell and HeLa cell underwent treatment with the extract at concentration ranging from 0 to 500μg/mL for 48 hours incubation period. The assessment of cell viability and determination of IC50 values were conducted utilizing the MTT assay. Visualization of apoptosis patterns was achieved through AO/PI staining followed by microscopic analysis, with quantification analysis by ImageJ software. The findings revealed a cytotoxic effect of the extract, as evidenced by IC50 values of 97,28μg/mL on T47D cells and 13,30μg/mL on HeLa cells. Microscopic examination of cells under fluorescence microscopy indicated apoptosis percentages exceeding 50%. This underscores the promising potential of Java cardamom seed as anticancer agent, capable of decreasing cancer cell viability and inducing apoptosis in breast cancer and cervical cancer.
