Abstract

Intercellular mitochondrial transfer assumed as one of therapeutic mechanism of mesenchymal stem cells (MSC) against diseases caused by mitochondrial damage, one of which is cardiomyopathy induced doxorubicin. The aim of this study is to determine the role of intercellular mitochondrial transfer as protective mechanism of MSC. This ex vivo study used neonatal rat cardiac given doxorubicin and MSC by using whole organ culture method. Miro1 gene expression measured as indicator of mitochondrial intercellular transfer activity and p53 gene expression as indicator of cellular stress. The expression of p53 gene in the group given doxorubicin 20µM for 30 minutes increased significantly (p<0.05) compared to the control group. The addition of 1x105 or 1x106 wharthon's jelly (WJ)-MSC at the 10th minute of culture caused the p53 gene expression to be significantly lower (p<0.05) than the treatment group without addition of WJ-MSC. No significant correlation between Miro1 gene expression and p53 gene was found in the WJ-MSC group. The conclusion of this study is intercellular mitochondrial transfer has no significant role in the therapy mechanism of WJ-MSC without genetic engineering.
