Abstract

Biopriming is a seed priming technique using beneficial microorganisms. Soaking duration is a factor affecting the success of priming techniques. Research on "The Effect of Soaking Duration on Biopriming Using Bacillus subtilis and Trichoderma harzianum On Rice Var. Anak Daro" had been conducted from March to August 2023 at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Universitas Andalas, Padang. This study aimed to determine the effect of soaking duration and biopriming agents on the germination of rice var. Anak Daro, the viability of B. subtilis and T. harzianum after biopriming, and antagonistic tests against M. oryzae. This study used experimental methods with a completely randomized design (CRD) factorial pattern with 2 factors with 3 replications. The results showed that Bacillus subtilis and Trichoderma harzianum had a significant effect on maximum growth potential and germination, whereas soaking duration had a significant effect on maximum growth potential and vigor index. The viability of Bacillus subtilis and Trichoderma harzianum increased in biopriming with 24 hours and 48 hours soaking duration but Trichoderma harzianum did not increase at 72 hours soaking duration Antagonist test results showed that Bacillus subtilis was able to suppress the growth of M. oryzae by 32.27% (medium category) and Trichoderma harzianum by 57.59% (high category).
