Abstract

PENGARUH EKSTRAK TERPURIFIKASI DAUN SURIAN (Toona sureni (Bl) Merr) TERHADAP DISFUNGSI SEL ENDOTEL YANG DIINDUKSI PREDNISON DAN NaCl ABSTRAK Nitrogen Monoksida (NO) adalah faktor relaksasi yang disintesa dan dilepaskan oleh sel endotel. Berkurangnya produksi NO dapat disebabkan karena disfungsi sel endotel. Daun surian (Toona sureni (Bl) Merr) diketahui mengandung metil galat yang telah terbukti sebagai antioksidan. Daun surian juga telah terbukti dapat meningkatkan kadar NO pada keadaan hiperkolesterolemia. Penelitian ini menguji efek proteksi ekstrak terpurifikasi daun surian terhadap disfungsi sel endotel. Penginduksi disfungsi sel endotel yang digunakan adalah Prednison dan NaCl 3 %. Sebanyak 25 ekor tikus putih jantan berusia 2 - 3 bulan dengan berat badan 200 - 300 g digunakan sebagai hewan uji. Hewan uji dibagi menjadi 5 kelompok, yaitu kelompok kontrol positif (diinduksi dengan Prednison dan NaCl), kontrol negatif (hewan normal), dosis 5 mg/kg BB, dosis 10 mg/kg BB, dan dosis 20 mg/kg BB. Dosis ekstrak terpurifikasi diberikan secara oral satu jam setelah pemberian penginduksi Prednison - NaCl selama 21 hari. Pengaruh efek proteksi terhadap disfungsi sel endotel diamati dengan mengukur konsentrasi NO serum. Hasil penelitian menunjukkan kelompok dosis 5 mg/kg BB berbeda nyata (p < 0,05) dengan kelompok kontrol positif, sedangkan kelompok dosis 10 mg/kg BB, dan 20 mg/kg BB tidak berbeda nyata (p > 0,05) dengan kelompok kontrol positif. Ini menunjukkan bahwa ekstrak terpurifikasi daun surian pada dosis 5 mg/kg BB mampu memproteksi disfungsi sel endotel yang diinduksi Prednison dan NaCl. EFFECT OF PURIFIED EXTRACT OF SURIAN LEAVES (Toona sureni (Bl) Merr) ON THE ENDOTHELIAL CELL DYSFUNCTION INDUCED BY PREDNISONE AND NaCl ABSTRACT Nitric Oxide (NO) is an Endothelial-Derived Relaxing Factor (EDRF). Reduce NO production can be caused by endothelial cell dysfunction. Surian leaves (Toona sureni (Bl) Merr) are contain methyl galat which antioxidant effect. Surian leaves also shown to increase NO level in hypercholesterolemic conditions. This study examined the protection effect of purified extract of surian surian leaves on endothelial cell dysfunction. Endothelial cell dysfunction induced by Prednisone and NaCl 3 %. A number of 25 white male rats aged 2-3 months, weighing of 200 - 300 g were used as experimental animals. The rats were divided into 5 groups, which consisted of positive Control (induced by Prednisone and NaCl), negative control (normal animal), dose 5 mg/kg BW, dose 10 mg/kg BW, and dose 20 mg/kg BW groups. Dosage of purified extract given orally one hour after induction of Prednisone-NaCl during 21 days. The protection effect on endothelial cell dysfunction was observed by measuring serum NO concentrations. The study showed that the group dose of 5 mg/kg BW was significantly different (p < 0.05) from the positive control group, while the group dose of 10 mg/kg BW, and 20 mg/kg BW was not significantly different (p > 0, 05) from the positive control group. This indicated that the purified extract of T. sureni at a doses of 5 mg/kg BW is able to protect endothelial cell dysfunction induced by Prednisone and NaCl.
