Abstract

“ISOLASI SENYAWA METABOLIT SEKUNDER DARI FRAKSI ETIL ASETAT DAUN TUMBUHAN “AKA LAMBUANG” (Merremia peltata (L.) Merr.,) DAN UJI AKTIVITAS ANTIBAKTERI“ ABSTRAK Merremia peltata (L.) Merr., merupakan salah satu tumbuhan yang digunakan sebagai obat tradisional. Penelitian ini bertujuan untuk mengisolasi senyawa metabolit sekunder dari fraksi etil asetat daun tumbuhan Merremia peltata (L.) Merr., dan uji aktivitas antibakteri. Isolasi senyawa dilakukan dengan metode ekstraksi dan metode kromatografi. Telah berhasil diisolasi tiga senyawa murni dengan notasi RL-14-16-5 (13, 2 mg), RL-14-27-2 (16 mg) dan RL-14-27-5 (10, 2 mg). Aktivitas antibakteri diuji dengan menggunakan metode dilusi. Masing-masing senyawa memberikan aktivitas sangat kuat terhadap 11 bakteri pathogen (Staphylococcus aureus ATCC 25922, Staphylococcus epidermidis ATCC 12228, Salmonella thypimurium ATCC 14028, Salmonella thyposa NCTC 786, Salmonella thypii, Bacillus subtilis ATCC 6633, Pseudomonas auruginosa ATCC 27853, Escherichia coli ATCC 11775, Micrococcus luteus ATCC 10240, Enterococcus faecalis ATCC 1054 dan Vibrio cholerae). Berdasarkan pemeriksaan kimia dan data spektroskopi, senyawa RL-14-16-5, RL-14-27-2 dan RL-14-27-5 merupakan senyawa golongan fenolik. Dari analisis GC-MS, senyawa RL-14-16-5, RL-14-27-2 dan RL-14-27-5 berturut-turut adalah 2-Propenoic acid, 3-(4hydroxy-3-methoxyphenyl), 4-Hydroxy-2-methylacetophenone dan Benzoic acid, 4-hydroxy. Merr.,) LEAVES” ABSTRACT Merremia peltata (L.) Merr., is one of plant used as traditional medicine. This study aims to isolate secondary metabolite compound from ethyl acetate fraction of Merremia peltata (L.) Merr., and to do the antibacterial activity test. The isolation was done by maseration, thin layer chromatography and colomn chomatography methods. Three pure compounds RL-14-16-5 (13.2 mg), RL-14-27-2 (16 mg) and RL-14-27-5 (10.2 mg) have been isolated. Antibacterial activity was tested using the dilution method. Each compound provides very strong activity against 11 pathogenic bacteria (Staphylococcus aureus ATCC 25922, Staphylococcus epidermidis ATCC 12228, Salmonella thypimurium ATCC 14028, Salmonella thyposa NCTC 786, Salmonella thypii, Bacillus subtilis ATCC 6633, Pseudomonas auruginosa ATCC 27853, Escherichia coli ATCC 11775, Micrococcus luteus ATCC 10240, Enterococcus faecalis ATCC 1054 and Vibrio cholerae). Based on chemical reactions and spectroscopic data, compounds RL-14-16-5, RL-14-27-2 and RL-14-27-5 are phenolic groups. Analysis by GC-MS, compounds RL-14-16-5, RL-14-27-2 and RL-14-27-5 are 2-Propenoic acid, 3- (4hydroxy-3-methoxyphenyl), 4 -Hydroxy-2-methylacetophenone and Benzoic acid, 4-hydroxy.
