Abstract
Introduction: Retinopathy of prematurity (ROP) in preterm infants is the leading cause of blindness in childhood. The method of oxygen-induced retinopathy in rats can be investigated for pathogcnesis and therapeutic intervention for retinal neovascularization in ROP. Retinal over-oxygenation plays an important role in ROP pathogenesis with rising ROS levels. L-carnitine (LC) has protective effects on tissues with some mechanisms against peroxidative stress by preventing the formation of ROS. Purpose: To assess the effects of L-carnitine (LC) on rats with oxygen-induced retinopathy to neovascular tuft formation. Method: The study was conducted on 36 Wistar rat pups. The rat pups were randomly divided into 2 groups. The first group (n=18) was exposed to hyperoxygen 75% (at 7-12 day postnatal) and received 0,2 mg/gram/day LC intraperitoneally (at 6-12 day postnatal). The second group 2 (n=18) was exposed to hyperoxygen 75% (at 7-12 day postnatal) and did not recieve LC. Both group were transferred to room air at 13 days postnatal. After postnatal day 20, the rat pups were killed and a histological examination was performed on the eyes, to investigate the neovascular tuft formation. Immunohistochemistry Griffonia simplicifolia lectin (GSL) has been developed to assess the proliferative neovascular response, quantified by counting the vascular endothelial nuclei that extend through the internal limiting membrane (ILM) towards the vitreous. This result is the average value of the neovascular tuft per 10-4 μm length of the retinal cross section. Result: The average body weight of rats in OIR group with LC was heavier than group without LC. There was a mean of 62,98 ± 14 neovascular tuft in OIR group without L-carnitine compared to 22,43 ± 9,87 neovascular tuft in OIR group with L-carnitine (p<0,05). Conclusion: LC has beneficial effects on oxygen-induced retinopathy in rats in terms of histopathological changes . Key Word: OIR, neovascular tuft, L-carnitine
