Abstract
Cervical cancer is a cancer attacked women and is responsible for the high mortality rate due to cancer in young women in developing countries. Cervical cancer is the second most common cancer that causing death in women with incidence rates in developing countries ranging from 40-100 per 100,000 people. Human Papillomavirus (HPV) is suspected as the leading cause of cervical cancer. HPV type-16 is classified as high-risk HPV that responsible in the majority of cases of cervical cancer. Identification of HPV genes in molecular biology techniques can be used as an early screening for cervical cancer. In this study, the samples used consisted of tissue samples and cervical smear samples of 43 samples obtained from RSUP. Dr. M. Djamil Padang and RSUD Arifin Achmad Pekanbaru. The method used is Polymerase Chain Reaction (PCR) method with GP5+/GP6+ primer and MY09+/MY11+ primer to identify DNA of HPV. The DNA amplification result using GP5+/GP6+ primer showed that 79.07% of the samples were infected by DNA HPV while the amplification result using the MY09+/MY11+ primer showed 34.88% of samples containing DNA HPV. Identification of genes E6, L1 and L2 HPV type-16 was performed using specific primers of E6, L1 and L2 genes HPV type-16. The results of identification showed that 46.51% of the samples contained E6 gene of HPV type-16, 30.23% of the samples contained L1 gene of HPV type-16 and 23.25% of the sample contained L2 gene of HPV type-16.
