Abstract

ABSTRAK Hiperurisemia merupakan salah satu penyakit yang tidak dapat diabaikan dalam masa kehamilan.Pada penelitian ini dilakukan uji terhadap pengaruh pemberian ekstrak etil asetat biji jintan hitam (Nigella sativa L) terhadap efek teratogen fetus mencit hiperurisemia. Hewan percobaan dibagi menjadi lima kelompok dimana masing-masing kelompok terdiri atas lima ekor hewan percobaan. Kelompok pertama merupakan kontrol yang diberikan aquadest. Kelompok kedua diberikan jus hati ayam dengan dosis 25 g/kgBB peroral. Kelompok tiga, empat dan lima diberikan ekstrak etil asetat biji jintan hitam dengan dosis berturut-turut 200 mg/kgBB, 400mg/kgBB dan 800 mg/kgBB setelah pemberian jus hati ayam 25 g/kgBB. Hasilpenelitian menunjukan bahwapemberian jus hati ayam 25 g/kgBB menyebabkan kelainan pada tulang ekor, fetus mati, hambatan pertumbuhan dan hemoragi. Pada pemberian jus hati ayam dan ekstrak biji jintan hitam 200 mg/kgBB menyebabkan hambatan pertumbuhan dan hemoragi. Pada pemberian jus hati ayam dan ekstrak biji jintan hitam 400 mg/kgBB menyebabkan kelainan pada tulang ekor, hambatan pertumbuhan dan hemoragi. Pada pemberianjus hati ayam dan ekstrak biji jintan hitam 800 mg/kgBB menyebabkan pendarahan, kelainan pada tulang ekor, hambatan pertumbuhan, fetus mati dan hemoragi. Berdasarkan uji analisis ANOVA didapatkan hasil bahwa pemberian ekstrak etil asetat tidak mempengaruhi berat badan induk, jumlah fetus dan berat badan fetus mencit hiperurisemia secara bermakna. Pemberian ekstrak etil asetat biji jintan hitam tidak berpotensi menghambat efek teratogen pada fetus yang mengalami hiperurisemia. Kata kunci: hiperurisemia, teratogen, biji jintan hitam, etil asetat ABSTRACT Hyperuricemia is one of fatal diseases during pregnancy. In this research, the teratogenic effect of black cumin (Nigella sativa L) ethyl acetate extract toward hyperuricemia mice fetus was examined. The experimental animals were divided into five groups. Each group consists of five experimental animals. The first, and second group were treated as a control group inducted with purified water, and orally induction with chicken liver juice at a dose of 25 g/kgBW, recpectively. Group three, four, and five were inducted using black cumin ethyl acetate extracts at various dose of 200, 400, and 800 mg/kgBW, after administration of chicken liver juice at dose of 25 g/kg BW. Results showed that the administration of chicken liver juice at a dose of 25 g/kgBW caused abnormalities of the coccyx, death in the fetus, an difficulties in growth and hemorrhage. The administration of chicken liver juice and black cumin seed extract at a dose of 200 mg/kgBW caused an difficulties in growth and hemorrhage. The administration of chicken liver juice and black cumin seed extract at a dose of 400 mg/kgBW caused acute an abnormality in the coccyx, difficulties in growth and hemorrhage, as well. The concomitant administration of chicken liver juice and 800 mg/kgBW of black cumin seeds extract caused bleeding, abnormalities in the coccyx, difficulties in growth, dead in fetus and hemorrhage. The ethyl acetate extract could not affectedsignificantlythe parent's body weight's, fetal numbers, and fetal body weight of hyperuricemia mice (p<0,05). In conclusion, ethyl acetate extract of black cumin seeds could not inhibit the teratogenic effect on a mice fetus with hyperuricemia.
