Abstract
IDENTIFIKASI DNA BETA SATELIT ISOLAT GEMINIVIRUS PENGINFEKSI TANAMAN CABAI (Capsicum annuum L.) DI PROVINSI SUMATERA BARAT ABSTRAK Penelitian dengan judul ’’Identifikasi DNA Beta Satelit Isolat Geminivirus Penginfeksi Tanaman Cabai (Capsicum annuum L.) di Provinsi Sumatera Barat’’ telah dilakukan di Laboratorium Bioteknologi dan Pemuliaan Tanaman Jurursan Budidaya Pertanian Fakultas Pertanian Universitas Andalas Padang dari bulan Juni sampai bulan Agustus 2012. Penelitian ini bertujuan untuk memetakan sebaran geminivirus monopartit pada pertanaman cabai di Provinsi Sumatera Barat dan melihat persamaan serta variasi sekuens DNA beta satelit di dalam populasi geminivirus yang ada di Provinsi Sumatera Barat. Isolat sampel geminivirus hasil isolasi dan hasil penelitian Jamsari et al. (2007) dari 6 wilayah Kabupaten/Kota di Provinsi Sumatera Barat (Pesisir Selatan dan Pasaman Barat mewakili dataran rendah, Solok dan Payakumbuh mewakili dataran sedang, selanjutnya Agam dan Tanah Datar mewakili dataran tinggi). Sampel tanaman cabai yang diindikasikan terserang geminivirus diisolasi dengan menggunakan metode Dellaporta et al. (1983) yang telah dimodifikasi. Kegiatan dilanjutkan dengan pengujian kualitas dan kuantitas DNA hasil isolasi dan penyetaraan konsentrasi. Amplifikasi dilakukan dengan menggunakan primer β01 -forward 5’-GGTACCACTACGCTACGCAGCAGCC- 3’ dan β02 – reverse 5’-GGTACCTACCCTCCCAGGGGTACAC- 3’. Sekuensing dilakukan di Laboratorium Dasar Universitas Andalas Padang secara satu arah (one read direction) menggunakan primer β01 - forward. Hasil sekuensing dianalisis menggunakan program BLAST untuk membandingkan sekuens yang diperoleh dengan sekuens yang telah didepositkan kedalam database (gen bank). Untuk melihat persamaan dan variasi sekuens DNA beta satelit dengan salah satu gen lain yang diperoleh dari hasil BLAST secara terperinci dilakukan penjajaran sekuens menggunakan program ClustalW2 secara online. Hasil penelitian menyimpulkan bahwa geminivirus monopartit yang mengandung beta satelit tersebar diseluruh areal pertanaman cabai di Provinsi Sumatera Barat (dataran tinggi = 55 %, dataran sedang = 50,8 % dan dataran rendah = 70 %). Tingkat kesamaan geminivirus yang mengandung beta satelit di Provinsi Sumatera barat sangat tinggi. Kata kunci : Identifikasi, Beta Satelit, Geminivirus, Cabai. IDENTIFICATION OF BETA SATELLITE ISOLATE DNA GEMINIVIRUS INFECTION PERFORMER FOR CHILI PLANTS (Capsicum annuum L.) IN WEST SUMATRA PROVINCE ABSTRACT Research with a title “IDENTIFICATION OF BETA SATELLITE ISOLATE DNA GEMINIVIRUS INFECTION PERFORMER FOR CHILI PLANTS (Capsicum annuum L.) IN WEST SUMATRA PROVINCE” has been done in Laboratory of Biotechnology and Plant Breeding, Department of Agriculture, Faculty of Agriculture, Andalas University Padang from June 2012 until August 2012. This research aimed for the distribution of geminivirus Monopartit on chili crop to be mapped in West Sumatera Province andsee the similarities and variations of beta satellite DNA sequences in the population geminivirus's in West Sumatra. Geminivirus isolated samples and research results Jamsari et al. (2007) of 6 regency / city in the province of West Sumatra (South Coast and West Pasaman represent low lands, Solok and Payakumbuh represent middle latitudes, Agam and Tanah Datar further represent plateau). Samples indicated chili plants infected geminivirus isolated using the method of Dellaporta et al. (1983) which has been modified. Activities continued with testing the quality and quantity of DNA isolation and concentration equalization. Amplification was performed using primary β01 - forward 5’-GGTACCACTACGCTACGCAGCAGCC-3’ and β02 – reverse 5’-GGTACCTACCCTCCCAGGGGTACAC-3’. Sequencing was performed in a basic laboratory Andalas University Padang with one read direction using primary β01 - forward. Sequencing results were analyzed using the BLAST program to compare the sequences obtained with sequences that have been deposited in to the database (bank gene). To see the similarities and variations of beta satellite DNA sequences with one of the other genes derived from the results of detailed BLAST sequence alignment performed using the program online ClustalW2. The research concludes that geminivirus monopartit containing beta satellite chili spread through out the planting area in the province of West Sumatra (plateau = 55%, moderate = 50.8% plains and low lands = 70%). Geminivirus similarities containing beta satellite in the province of West Sumatra is very high. Key Words : Identification, Beta Sattelite, Geminivirus, Chili
