Abstract
ABSTRAK Antibiotik merupakan metabolit sekunder yang dihasilkan oleh mikroba terutama jamur. Peneliti terdahulu telah berhasil menapis empat jenis jamur yang hidup disarang ratu termite Macrotermes gilvus Hagen., salah satunya Aspergillus flavus. Jamur ini bertahan menghasilkan senyawa metabolit pada media SDA hanya pada ekstrak sub-kultur biakan pertama dan kedua, senyawa metabolit berangsur hilang pada biakan berikutnya. Hal ini diduga terjadi karena perbedaan habitat tumbuh, oleh karena itu dilakukan pengayaan media dengan tanah sarang ratu termite yang bertujuan untuk mendapatkan kembali metabolit awal. Pengayaan dilakukan menggunakaan empat konsentrasi media berbeda (0,25; 0,50; 0,75 dan 1 gram sarang/mL media). Hasil yang diperoleh menunjukkan pengayaan media SDA dengan 1 gram sarang/mL media memberikan pertumbuhan jamur paling optimal. Jamur sub-kultur biakan ketiga dibiakkan pada media dan menjadi biakan keempat. jamur dibiakkan hingga sub-kultur biakan kesepuluh, dilakukan ekstraksi dan fraksinasi terhadap masing-masing biakan jamur. Berdasarkan analisis profil KLT fraksi etil asetat jamur, senyawa metabolit awal belum terbentuk hingga sub-kultur kesepuluh, namun terbentuk 6 spot noda baru golongan terpenoid. Hasil kromatografi kolom didapatkan 10 sub-fraksi. Uji aktivitas antibiotik dilakukan dengan metode difusi terhadap 12 bakteri dan 3 jamur uji. Spot noda 2, 5, dan 6 (Rf 0,84; 0,36; 0,26) dapat menghambat pertumbuhan bakteri Pseudomonas aeruginosa ATCC 27853, spot noda 3, dan 4 (Rf 0,74; 0,52) dapat menghambat pertumbuhan bakteri Micrococcus luteus ATCC 10240. ABSTRACT Antibiotic is secondary metabolites produce by microbes especially fungus. Previous study had succesfully screened four types of fungus that lived in queen termites’s nest, one of them was Aspergillus flavus. This fungus lasted to produce metabolites compound on SDA media which only existed in first and second sub-culture extract, faded gradually on the next culture. It was presumably happened because of a different habitat to grow. Therefore, it was necessary to enriched the media with queen termite nest to get back the initial metabolites. The enrichment was done by using four different media concentrations (0.25; 0.50; 0.75 and 1 gram of nest/mL media). The result showed that enrichment of 1 gram of nest/mL media provided the most optimum fungus growht. The third sub-culture of Aspergillus flavus was bred on enriched media which would become the fourth sub-culture, this fungus was bred until the tenth sub-culture, and then the extraction and fracination to each culture was done. Based on TLC’s profile analysis, the initial metabolites didn’t formed until the tenth sub-culture yet, but six new spots terpenoid compounds were formed. The result of column chromatography obtained 10 sub-fraction. The activity test of antibiotic was done by diffusion method against 12 test bacterias and 3 fungus test. Stain spot 2, 5, 6 (Rf 0.84; 0.36; 0.26) could inhibit the growht of Pseudomonas aeruginosa ATCC 27853, stain spot 3 and 4 (Rf 0.74; 0.52) inhibit the growht of Micrococcus luteus ATCC 10240.
