Abstract

The mutation of PIK3CA gene altered the activation of phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K) in controlling cell proliferation and induced cancer cell formation. Aim of this study is to determine exon 9 mutation of PIK3CA gene in breast cancer by using PCR method and Sanger sequencing method. Genomic DNA was isolated from ten breast carcinomas. Exon 9 of PIK3CA gene was amplified by using specific primer. Furthermore, the purified PCR products were sequenced. The exon 9 of PIK3CA gene sequences was analyzed by Geneious software. The isolated genomics DNA with concentration between 80-140 ng/µL were used to amplify 321 bp PCR products harbor exon 9 of PIK3CA at annealing temperature 59 °C. The exon 9 of PIK3CA gene sequences in tumor samples showed 100% similarity with the NCBI reference sequences of PIK3CA gene. The designed primer successfully amplified region for exon 9 of PIK3CA gene although no mutation found in exon 9 of PIK3CA gene in ten breast cancer patients.
