Abstract

THE EFFECT OF VITAMIN E SUPPLEMENTATION TOWARDS THE LEUKOCYTES AND NEUTROPHILS COUNT; AN EXPERIMENTAL STUDY USING ACUTE CIGARETTE SMOKE EXPOSED MICE By KHOIRUN NISA ABSTRACT Acute cigarette smoke exposure contains 1014 free radicals/puff that cause oxidative stress. Stress oxidative initiate lipid peroxidation and endothelial cell dysfunction that induce inflammatory response, as marked by elevated of leukocytes and neutrophils counts. Vitamin E as an antioxidant, protect cells membrane from oxidation of free radical. This research aims to determine the effects of vitamin E on leukocytes and neutrophils counts were given acute cigarette smoke exposure in mice. Research Randomized Post Test Only Control Group Design conducted on 24 male mice were divided into four groups: negative control, positive control, P1 and P2. Positive control group was given acute cigarette smoke exposure two cigarette twice a day for three days, P1 and P2 groups were given vitamin E containing 0,26 IU/20gBB and 1,5 IU/20gBB for 10 days. Further, P1 and P2 groups were given acute cigarette smoke exposure two cigarette twice a day for three days. Based on the results of statistical tests shows that there is significant differences between positive control group and negative control groups, positive control group and P1 group, positive control group and P2 group, while P1 group and P2 group had no significant difference. This study concludes that vitamin E can influence on leukocytes and neutrophils counts was given acute cigarette smoke exposure. Keywords : vitamin E, acute cigarette smoke, leukocytes and neutrophils counts PENGARUH PEMBERIAN VITAMIN E TERHADAP JUMLAH LEUKOSIT DAN NEUTROFIL MENCIT YANG DIPAPAR ASAP ROKOK AKUT Oleh KHOIRUN NISA ABSTRAK Paparan asap rokok akut yang mengandung 1014 molekul radikal bebas dalam sekali hisapan dapat menyebabkan terjadinya stres oksidatif. Stres oksidatif mengakibatkan peroksidasi lipid dan kerusakan sel endotel yang memicu reaksi inflamasi, ditandai dengan terjadinya peningkatan jumlah leukosit dan neutrofil. Vitamin E sebagai antioksidan, melindungi membran sel dari oksidasi radikal bebas. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian vitamin E terhadap jumlah leukosit dan neutrofil mencit yang dipapar asap rokok akut. Penelitan Randomized Post Test Only Control Group Design dilakukan pada 24 mencit jantan yang dibagi dalam 4 kelompok: kontrol negatif, kontrol positif, P1 dan P2. Kelompok kontrol positif diberikan paparan asap rokok akut 2 batang 2 kali sehari selama 3 hari, kelompok P1 dan P2 diberikan vitamin E masing-masing 0,26 IU/20gBB dan 1,5 IU/20gBB selama 10 hari selanjutnya diberikan paparan asap rokok akut selama 3 hari. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan antara kelompok kontrol positif dengan kelompok kontrol negatif, kelompok kontrol positif dengan kelompok P1, dan kelompok kontrol positif dengan kelompok P2. Namun tidak terdapat perbedaan yang signifikan antara kelompok P1 dan kelompok P2. Kesimpulan penelitian ini adalah pemberian vitamin E berpengaruh terhadap jumlah leukosit dan neutrofil yang dipapar asap rokok akut.
