Abstract

Cervical cancer caused by high-risk Human Papillomavirus (HPV). Cancer will secrete Tumor Necrosis Factor Alpha (TNF-α) to create a microtumor environment supportive of cancer survival. The common therapies for cervical cancer are surgery, radiotherapy, chemotherapy, and medicinal plants, one of which is sungkai. Sungkai (Peronema canescens J) leaf ethanol extract contains compounds that act as anti-inflammatories and antioxidants. This study aimed to determine the administration of Sungkai leaf ethanol extract in reducing TNF-α gene expression in HeLa cells. This study used 24 well plates of HeLa cell culture that grew confluently and were divided into four groups (K, P1, P2, and P3). The treated group was administered with Sungkai leaf ethanol extract with concentrations of IC25, IC50, and IC75 which were incubated for 24 hours. The TNF-α gene was examined using conventional PCR, and then the bands were analyzed using ImageJ. Based on the results of data analysis using One Way ANOVA and Post Hoc Bonferroni tests, the average TNF-α gene expression in groups K, P1, P2, and P3 were respectively 0,32; 0,29; 0,23; and 0,14. There was a significant difference between group K to P2 and P3 with a value of p=0.000 (p<0.05), while the difference between group K and P1 was not significant (p>0.05). The conclusion obtained was that TNF-α gene expression was significantly decreased in the HeLa cells group treated with concentrations IC50 and IC75 of Sungkai (Peronema canescens J) leaf ethanol extract.
