Abstract
Exposure of lead acetate as free radicals can damage cells, it can increase the compound lipid peroxide (LPO). LPO levels can be detected through one of its stable product, malondialdehyde (MDA). The body’s effort to neutralize the LPO need a lot of antioxidants. Vitamin C is a water soluble antioxidant that can inhibit the formation of LPO. The objective of this study were to determine the effect of vitamin C on MDA serum level of white wistar strain rats were exposed to lead acetate. This study was used true experimental randomized control group post test only design and conducted at Pharmacology Laboratory of Pharmacy Faculty and Biochemistry Laboratory of Medical Faculty Andalas University held from May 2016 until November 2016. The study was conducted for four weeks for 24 wistar rats which divided into four groups consisting of : (C-) negative control, (C+) were exposed lead acetate 0.05 mg/ g bw/ day, T1 were exposed lead acetate 0.05 mg/ g bw/ day and given vitamin C doses of 0.05 mg/ g bw/ day and T2 were exposed lead acetate 0.05 mg/ g bw/ day and given vitamin C doses of 0.075 mg/ g bw/ day. The result showed that the malondialdehyde serum levels at (K-) and (K+) are 2.94 nmol/ml and 3.94 nmol/ml, (P1) and (P2) are 3.3 nmol/ml and 2.95 nmol/ml. The differences of MDA serum levels between groups by ANOVA p = 0.000. It could be concluded that vitamin C in doses 0,05 mg/ g bw/ day and 0,075 mg/ g bw/ day have significantly affect to decrease the MDA serum levels of white rats wistar strain that were exposed to lead acetate. Keywords :Vitamin C, Malondialdehyde serum, Lead acetate EFEK PEMBERIAN VITAMIN C TERHADAP KADAR MALONDIALDEHID SERUM TIKUS PUTIH GALUR WISTAR YANG DIPAPARKAN PLUMBUM ASETAT Oleh WIDYA DWI JAMILZA ABSTRAK Pemaparan plumbum asetat sebagai radikal bebas dapat merusak sel, hal ini meningkatkan senyawa lipid peroksida (LPO). Kadar LPO diketahui melalui salah satu produknya yang stabil yaitu malondialdehid (MDA). Usaha tubuh untuk menetralisir LPO membutuhkan antioksidan yang banyak. Vitamin C merupakan salah satu antioksidan yang larut dalam air yang mampu menghambat pembentukan LPO. Penelitian ini bertujuan untuk melihat efek pemberian vitamin C terhadap kadar malondialdehid serum tikus putih galur wistar yang dipaparkan plumbum asetat. Rancangan penelitian adalah eksperimental murni randomized control group post test only design yang dilakukan di Laboratorium Farmakologi Fakultas Farmasi dan Laboratorium Biokimia Fakultas Kedokteran Universitas Andalas pada bulan Mei 2016 hingga November 2016. Penelitian dilakukan selama empat minggu pada 24 ekor tikus galur wistar yang dibagi menjadi empat kelompok yang terdiri dari (K-) kontrol negatif, (K+) dipaparkan Pb asetat 0,05 mg/ g BB/ hari, (P1) dipaparkan Pb asetat 0,05 mg/ g BB/ haridengan vitamin C dosis 0,05 mg/ g BB/ hari dan (P2) dipaparkan Pb asetat 0,05 mg/ g BB/ hari dengan vitamin C dosis 0,075 mg/g BB/ hari. Hasil penelitian menunjukkan kadar MDA serum pada (K-) dan (K+) : 2,94 nmol/ml dan 3,94 nmol/ml, (P1) dan (P2) 3,3 nmol/ml dan 2,95 nmol/ml. Perbedaan kadar MDA serum antar kelompok dengan uji ANOVA p= 0,000. Kesimpulan penelitian bahwa pemberian vitamin C 0,05 mg/ g BB/ hari dan 0,075 mg/ g BB/ hari memberikan efek yang signifikan dalam menurunkan kadar MDA serum tikus putih galur wistar yang dipaparkan plumbum asetat.
