Abstract
Microalgae Isolation from Maninjau Lake West Sumatra Potentially as Antibacterial Microalgae are microorganisms photosynthetic prokaryotic and eukaryotic produced bioactive compounds can be used as antibacterial agent. In this research, isolation of microalgae from Maninjau Lake West Sumatra was identified by morphological and molecular identification by PCR using primers 16S rDNA prokariotic microalgae and 18S rDNA to eukaryotic microalgae. Tests on the cover isolates the growth curve, the weight of dry biomass and inhibition of the extract using a solvent n-hexane and methanol, phytochemical analysis and antibacterial activity against pathogenic bacteria as Escherichia coli, Staphylococcus aureus, Bacillus subtilis and Bacillus cereus using method disc diffusion. The results of the study, there are three microalgae isolates identified that isolates AUMA-020 (Scenedesmus), isolates AUMA-23 (Unculture cyanobacterium) and AUMA-019 (Unculture Limnothrix sp.). Isolates AUMA-20 and isolates AUMA-23 experiencing late stationary phase on day 21 with the weight of the dry biomass of 69.2% and 83.70%. Each isolate was extracted with a solvent n-hexane and methanol extracts with a yield of 7.8%, 18.15% and 5.06% and 22.38%. To these two isolates of the methanol extract shows that it contains phenolic compounds, flavonoids, terpenoids and steroids, while the extract of n-hexane isolates AUMA-020 containing compounds terpenoids and saponins and isolates AUMA-023 only compounds containing terpenoids. The test results of antibacterial activity of extracts of n-hexane and the methanol extract of the test bacteria Staphylococcus aureus, Bacillus cereus and Bacillus subtilis clear zone, respectively for 25, 20, 21 mm and 11, 11, 12 mm (isolates AUMA-020) and 24.5, 16, 17.5 mm and 9.5, 9, 10 mm (isolates AUMA-023). Both isolates were equally did not show any antibacterial activity against Escherichia coli. It can be concluded the antibacterial activity of two isolates of microalgae extract n-hexane is better than the methanol extract and isolate microalgae isolated from in Maninjau Lake West Sumatera potential as an antibacterial agent.
