Abstract
The isolation of secodary metabolites from Miana leaves has been completed. The isolation was carried out applying both maseration method with the use of methanol as a solvent and fractination methodxusing solvent hexane and ethyl acetate. Antioxidant of fractions of each solvent was tested by using free radical method 1,1-diphenyl-2-picrylhydrazyl and IC50 values of hexane, ehtyl acetate and methanol fraction assesed at 141,86 ; 20,52; & 24,76 mg/L respectively. Those data indicate that ethyl acetate fraction of miana leaves is the most active antioxidant with IC50 20,52 mg/L. Then ethyl acetate fraction was chromatographed employing silica gel as stationary phase whilst hexane, ethyl acetate, and methanol solvent as mobile phase with Step Gradien Polarity (SGP) system. Isolation yield compound belong in flavonoid group in the form of white solid with Rf 0,7 (ethyl acetate : methanol 6 : 4) and provides a single blue stain under the beam of UV 365 nm light.
