Abstract
Grammatophyllum stapeliiflorum is listed in appendix II of the CITES. The germination of the orchid in nature is low and depend on mycorrhiza fungi to provide nutrients. Tissue culture technique is one of solution for propagating of orchide using seed as source of explants. Pepton is an organic compound that can increase the germination and protocorm development because containing amino acid, biotin protein, pyridoxine, thiamine and nitrogen. The aim of the research were to determine the effect of pepton on seed germination and protocorm development and also to determine the best concentration peptone for seed germination and protocorm development of G. stapeliiflorum by tissue culture technique. The research used a Completely Randomized Design (CRD) with 5 treatments and 6 replications. The mature seed were sown on MS medium and added with various concentrations of peptone (0.5; 1; 1.5; 2; and 2.5 g/L) and without peptone as a controle treatment. Observation of the protocorm formation was carried out every day until the first protocorm was formed. The data analiyzed statistically using the one-way Anova test to identify the effect of pepton and DNMRT test to identify the significance of each treatment. The stage of the embryo development was observed for 10 weeks after sown and analyzed descriptively. The results of statistical analysis showed that peptone had a significant effect in accelerating germination. The addition of 2 g/L peptone in MS medium was the suitable concentration for seed germination (88.59%), protocorm formation (61.02%) and protocorm with SAM and RAM (22.43%).
